Classic concepts have assumed that β3-AR agonists 23 work by directly acting on a β3-AR located in the plasma membrane of smooth muscle cells in 24 the detrusor of the urinary bladder (15). Recently, this concept has been critiqued increasingly 25 (7) because mirabegron, the only β3-AR agonist currently in clinical use, has an EC50 for 26 Articles in PresS. On the Site and Mechanism of Action of β3-Adrenoceptor Agonists in the Bladder

The clinical success of mirabegron as the first β3-adrenoceptor (AR) agonist for treatment of the overactive bladder (OAB) syndrome, has resulted in substantial interest in its site and mechanism of action. Even if the adrenergic innervation of the bladder and urethra has been well studied, the location(s) of β3-ARs in different structures within the bladder wall and urethra, and the mode(s) of...

Can [125I]-Iodocyanopindolol Label β3-Adrenoceptors in Rat Urinary Bladder?

β(3)-Adrenoceptors have been demonstrated to mediate urinary bladder smooth muscle relaxation but proof of their expression at the protein level has been missing because of lack of suitable antibodies or radioligands. As among various available radioligands [(125)I]-iodocyanopindolol ([(125)I]-ICYP) exhibited the smallest problems in labeling cloned human β(3)-adrenoceptors in previous studies,...

Which -Adrenoceptor Subtypes are Important in the Treatment of Over- active Bladder?

For the pharmacological treatment of frequency, urgency and/or urge incontinence due to bladder overactivity, anticholinergics have been used as first-choice drugs. However, these drugs can exert severe side effects (accommodation paralysis, constipation, tachycardia, dry mouth and blurred vision) and some patients are refractory to their actions. Thus there is a need for new drug therapies wit...

Muscarinic receptor subtypes of the bladder and gastrointestinal tract.

The parasympathetic nervous system is responsible for maintaining normal intestinal and bladder function, contracting the smooth muscle by releasing the neurotransmitters acetylcholine (ACh) and ATP and relaxing sphincters by releasing nitric oxide. ACh is the main transmitter released and smooth muscle contraction is mediated via a mixed M2/M3 receptor population; M3 receptors acting via phosp...

Beta3-Adrenoceptor Agonists: Possible Role in the Treatment of Overactive Bladder

In the present review article, we present an overview of beta-adrenoceptor (β-AR) subtype expression at the mRNA and receptor protein levels in the human detrusor, the in vitro and in vivo bladder function of the β3-AR, the in vivo effect of β3-AR agonists on detrusor overactivity in animal models, and the available results of clinical trials of β3-AR agonists for treating overactive bladder (O...
